摘    要:用野生型发根农杆菌(Agrobacterium rhizogenes)A4菌株感染骆驼刺(Alhagi pseudoalhagi)无菌苗的子叶和下胚轴,15d左右在感染部位诱导出发根。将发根切段放在附加2.0mg/L2,4-D和0.5mg/L6BA的MS培养基上诱导愈伤组织,在3.0mg/L6BA的MS培养基上进一步分化形成完整植株,冠瘿碱检测结果表明,不仅在初生发根中,在次生发根、来源于发率的愈

Abstract:By using Agrobacterium rhizogenes A4with Ri- plasmid,the transformation test was carried out with the cotyledons and hypocotyls of Alhagi pseudoalhadi.Transgenic plants of Alhagi pseudoalhagi were regenerated from aseptic calluses derived from hairy roots transformed by Agrobacterium rhizogenesstrains A4at or near the site of infection of cotyledon and hypocotyl segments.Both agropine and mannopine were detected not only in extract of the primary hairy roots,but also in those of the secondary hairy roots,callus de- rived from hairy roots and leaves of regenerated plantlets.Compared with hypocotyl seg- ments,the time of hairy root induction from cotyledon segments was shorter and the fre- quency of transformation was higher.
