小粒径PLGA纳米颗粒的制备

摘    要:聚乳酸—羟基乙酸共聚物(PLGA)因具有良好的生物相容性,已成为发展最好的可降解聚合物之一,而其粒径是限制其应用的一个重要因素.通过双乳化-溶剂挥发(W/O/W)方法制备小粒径PLGA纳米颗粒.并用纳米粒度及电位分析仪(DLS)对其粒径及Zeta电位进行研究讨论.结果表明,不同的水油比,表面活性剂浓度以及有机相浓度对粒径都有影响.在水油相比大于16∶1,PLGA与表面活性剂质量比大于1∶1,有机相浓度为5 mg/m L时,都出现了小于100 nm的PLGA纳米颗粒,所得PLGA纳米颗粒的粒径范围在49 nm~445 nm.

Abstract:Poly(latic acid-co-glycolic)(PLGA) has been intensively studied as one of biodegradable polymers due to its biocompatibility. Size control of the PLGA nanoparticles is important for the construction of drug delivery systems. In this paper,PLGA nanoparticles prepared by a double emulsion-solvent method is reported. Characterization of the nanoparticles is mainly performed by dynamic light scattering(DLS) measurement. It is revealed that the volume ratio of organic solvent and water,the content of surfactant,and the content of organic phase influence the particle size of the PLGA nanoparticles. In this work we have obtained the PLGA nanoparticles with particle size in the range of 49 nm ~ 445 nm.
