摘    要:研究了液相色谱-串联质谱法测定血浆中丹参素含量的方法.在实验确定的样品处理及液相色谱-质谱测定条件下,丹参素在2-400ng·mL。范围内呈良好的线性关系,加权最小二乘法计算得r为0.9985,最低定量限为2ng·mL^-1;血浆中加入丹参素浓度为5.0、50.0和400.0ng·mL“时的绝对回收率分别为54.3%、55.2%和51.6%,准确度(RE%)分别为-9.90%、-5.42%和4.50%,日内、日间精密度(RSD)分别为5.57%、5.54%、6.36%和2.86%、6.22%、9.02%.该方法灵敏度高、精密度和准确度好,满足药代动力学研究的要求.

Abstract:A sensitive liquid chromatography-tandem mass spectrometric (LC/MS/MS) method for assaying Danshensu in dog plasma was developed and fully validated. With the optimized LC/MS/MS and sample preparation method, the calibration curves show a good linearity over the range of 2 -400 ng·mL^-1. Fitted with a weighted linear least-squares regression analysis, the linear regression (r) is 0.9985. The method is validated at three concentrations of 5.0,50 and 400 ng·mL^-1 of Danshensu. The intra- and inter-day precision is 5.57%,5.54%, 6.36% and 2.86%, 6.22% 9.02%, the extract recovery is 54.3%, 55.2% and 51.6%, and the accuracy ( RE% ) is - 9.90%, - 5.42 % and 4.50 %, respectively. The assay is sensitive, accurate and suitable for pharmacokinetic study of Danshensu in plasma.
