摘    要:采用氨基酸分析仪建立了土鳖虫中谷氨酸含量的检测方法,比较不同浓度的盐酸以及不同料液比、提取时间对土鳖虫中谷氨酸提取效果的影响。结果表明,土鳖虫中谷氨酸的最佳提取工艺为6 mol/L 的盐酸消解,料液比1:1(mg/mL),水解时间22 h,建立的谷氨酸分析仪法回收率为99.2%,精密度试验相对标准偏差为0.19%,不同地区土鳖虫中谷氨酸质量分数存在较明显差异,范围为1.068%~1.149%。该方法重现性好、快速高效、操作简便,适用于土鳖虫中氨基酸含量的检测,可为其质量控制及资源深入开发提供技术支持。

Abstract:The effects of various concentrations ofhydrochloric acid, material-to-liquid ratios, and extraction times on theextraction of glutamate in Eupolyphaga Steleophaga were compared using an aminoacid analyzer to establish a method to measure the glutamate content inEupolyphaga Steleophaga. The results showed that the best extraction processfor glutamic acid in Eupolyphaga Steleophaga was a material-to-liquid ratio of1:1 (mg/mL), 6 mol/L hydrochloric acid, and hydrolysis time of 22 h. Therecovery rate of the glutamic acid analyzer method was 99.2%, and the relativestandard deviation of the precision test was 0.19%. There were significantdifferences in the glutamate content in Eupolyphaga Steleophaga from differentregions, ranging from 1.068% to 1.149%. The method is reproducible, fast,efficient, and easy to perform. It is suitable for the detection of amino acidcontent in Eupolyphaga Steleophaga and can provide technical support for itsquality control and in-depth resource development.
