摘    要:以羽化后未交尾的雌性麻蝇为活体材料,通过“Feulgan预染——显微解剖悬液铺片——Giemsa复染法”获得形态良好、分散适中、着色清晰的卵泡细胞有丝分裂中期染色体标本。本法材料性别来源明确,对蝇类性染色体鉴定的准确性有重要意义。

Abstract:This paper described a new method for preparing the ovary mitotic metaphase chromosomes in flies. The material used in this study was the female flesh-flies (Parasarcophaga similis) which did not mate after emerging. Each fly was injected with 0.02ml of 0.1% colchicine (W/V) and kept under the room temperature for about 1,5h. Then the ovaries were taken out from the flies. It was followed by placing the ovaries in 0.007% KCI for 1h, fixing in 3 : 1 methanol-acetic acid, and prestaining with Feulgan staining method. Then the ovarioles were picked out from the ovaries on a slide with a drop of 60% acetic acid under a stereomicroscope to make the follicle cells became cell suspension. Then some methanol-acetic acid (3 : 1)was dropped down to the suspension to spread the follicle cells and their chromosomes on the slide. Air-dried slides were restained with Giemsa. Thus, the clear and well-spreaded metaphase chromosomes of follicle cells were obtained. The new method mentioned above makes it possible to use female adult fly for preparing mitotic metaphase chromosomes directly and recognize sex chromosomes precisely.
