Ri质粒转化牛膝及其发状根的培养

摘    要:用发根农杆菌的Ri质粒转化药用植物牛膝,诱导出了抗卡那霉素的发状根。将诱导的发状根进行固体平板培养和液体悬浮培养。在附加Km30mg/L的MSo和1/2MSo培养基上,发状根生长良好,而未经转化的对照组则死亡,表现出生激素自主性。在附中外源激素IBA0.1mg/L和NAA0.1mg/L时,能促进发状根的成活,侧根的形成以及提高增殖倍数。表明适量的外源激素能促进发状根的生长。

Abstract:Achyranthes bidentata BL, a famous medica plant, was transferred by Ri plasmid with Agrobacterium rhizogenes , and haiy root with kanamycin resistance was induced successfully. The induced roots were cultured in solid media and liquid media. The excised hairy roots grew wery well on MSo and 12MSo media which both contained Km30mg/L, where as the control group can not. These results showed that the hairy root has the ability of anxin synthesis by itself. Media containing IBA0.1mg/L or NAA 0.1mg/L can increase the hairy root survival rate, lateral root formation and cultural production, which demonstrated that proper foreign auxin can promote the growth of hairy root.
