摘    要:借离子对反相高效液相色谱分离美满霉素(MNC)、土霉素(OTC)、美他霉素 (MTC)和金霉素(CTC)的铝(Ⅲ)螯合物并于激发波长380nm和发射波长480nm处 检测.研究了螯合物的最宜分离和测定条件.于KromasilODS柱(250×4.6mm I.D.,5μm)上,用含0.050mol/L柠檬酸缓冲剂(pH2.5)作流动相,流速为0.7mL/ min.检测限为0.12ngMNC,0.2ngOTC,1.8ngMTC和0.6ngCTC.

Abstract:Minocycline(MNC), oxytetracycline(OTC), methacycline(MTC) and chlortetracy cline(CTC) as chelates derivatized pre-column in a solution of 0.4 mol/L acetic acid-sodium acetate buffer (pH 4.0) with aluminium( Ⅲ ) were separated by ion-pair reversed-phase high performance liquid chromatography and detected spectrofluorimetrically with excitation wave length at 380 nm and emssion wavelength at 480 nn. The optimum conditions for the separation and determination of MNC-, OTC-, MTC- and CTC-AI( Ⅲ ) chelates were investigated. The chelates were separated on a Kromasil ODS column (250× 4.6 mm I.D., 5 μn) using ace tonitrile-dimethylfonnamide-water (5: 20: 75, V/V/V) containing 0.050 mol/L citric acid buffer (pH 2.5) as a mobile phase at a flow rate of 0.7 mL/min. The detection limits of the assay (signal-to-noise ratio = 3: 1) were 0.12 ng, 0.2 ng, 1.8 ng and 0.6 ng for MNC, OTC, MTC and CTC, respectively.
