^99mTc直接标记单克隆抗体3H11及其在小鼠体内分布的研究

摘    要:对二硫苏糖醇(DTT)为抗体还原剂,亚锡MDP药盒为弱交换配体,进行了^99mTc直接标记McAb3H11的研究。用HPLC分析鉴定标记物,标记率大于95%,标记后不需纯化。体外竞争实验和在小鼠体内分布的研究表明,由标记得到的^99mTc-3H11和^99mTc-IgG,在体内和体外均是稳定的。

Abstract:The direct labelling of McAb3H11 with 99m Tc using dithiothreitol (DTT) as reducing agent of antibody and stannous MDP kit as weak exchange ligand was investigated.Labelled 3H11 was analysed and identified by HPLC.The labelling efficiency was more than 95%.Hence no purification was needed.Competetion results in vitro and biodistribution in mice showed that 99m Tc 3H11 and 99m Tc IgG were stable in vitro and in vivo.
