经产大鼠肝郁气滞型乳腺增生病模型的建立

摘    要:目的建立经产大鼠乳腺增生病病证结合模型。方法雌性经产未孕SD大鼠隔2 d肌肉注射苯甲酸雌二醇注射液1.0 mg.kg^-1,连续10次后次日开始每日肌肉注射黄体酮5.0 mg.kg^-1,连续5 d;在造模过程中隔日将其装入束缚笼固定过夜,共15次。在末次注射黄体酮次日测量大鼠乳头直径,进行旷场实验;禁食12 h,速眠新麻醉股动脉取血备检,处死取乳腺备检。结果模型组大鼠较正常组乳头肿胀,旷场实验中格停留时间延长,大鼠穿行格数和站立修饰次数减少,站立修饰时间缩短。模型组大鼠血清泌乳素(PRL)、雌二醇(E2)升高,孕醇(P)降低,乳腺腺泡数量增多,体积增大,腺泡腔扩张,充满嗜碱性分泌物,模型大鼠各项指标3周内未恢复至正常水平。结论激素结合束缚方法成功建立经产大鼠肝郁气滞型乳腺增生病模型。该方法造模结果可靠,符合乳腺增生病临床发病病因,方法简单。

Abstract:Objective To establish a multiparous rat cyclomastopathy model.Methods Twenty multiparous female rats were divided into normal and model groups,and the latter were injected through muscular with oestradiol benzoate for ten times at a dose of 1.0 mg·kg-1 once every two days.From the last injection of oestradiol benzoate,luteosterone was given to rats for 5 days.In the whole procedure,rats were enclosed into fastening cages at night every two days for 15 times.After last injection of luteosterone,rats were sacrificed and galactophore were collected.Results The number of gland alveolus increased more obviously than that of the normal groups,the volume of gland alveolus inflated,the acinar luminas expanded and the wall of gland alveolus was thinner and filled fully with basophilia secretion.All of those changes didnt recover to normal levels in 3 weeks.Conclusion Multiparous rat cyclomastopathy model can be established by hormones induction combined with fastening method.According to our study,this method is reliable,effective and simple.
