摘    要:探讨人尿样标本保存方法对异黄酮测定结果的影响.利用高效液相色谱法对比分析人尿在不同解冻温度、不同冻融次数、不同放置时间对异黄酮(大豆苷原、染料木黄酮、黄豆黄素、雌马酚)测定结果的影响.检测分析结果表明,人尿在4℃、室温(25℃)和37℃条件下解冻对异黄酮组分的测定结果的影响不明显(P〉0.05);室温条件下一次冻融或多次冻融,各项检测结果仍比较稳定;尿液标本在4℃或室温放置24h内以及-20℃放置3周内对异黄酮测定结果影响不大.

Abstract:TO explore the effect of preservation conditions of human urine samples on detection of isoflavone. The effect of different frozen-thawing temperatures, frequencies, keeping time of human urine on its isoflavones (daidzein, glyeitein,equol, genistein) contents determination was investigated by using high performance liquid chromatography (HPLC)method. The present results showed that no significant differences were observed under 4 ℃, room temperature (RT) and 37 ℃. Under RT,stable detecting results of all of isoflavones were obtained with either frozen-thawing once or several times. It is concluded that isoflavones contents in human urine are very stable not only under 4 ℃ or room temperature (RT) for 24 h,but also under - 20 ℃ for 3 weeks.
