石刁柏花药培养中根、茎及胚状体的形态发生研究

摘    要:利用扫描电镜及组织学方法观察了石刁柏(Asparagus of ficinalis)的花药愈伤组织及由该愈伤组织产生的根、茎和胚状体。愈伤组织可分为三种,其表面形态及将来的发育方向都有差异。胚状体有内、外两种起源方式,胚状体的发生是不同步的,其发育顺序及结构与合子胚大体相似,有少数胚状体形成了植株。茎端最早是在愈伤组织表层产生,茎端具典型的原套原体结构,原套细胞仅一层。培养产生的根、茎与自然生长的根、茎在形态结构上有显著的差异。

Abstract:Using SEM and histological methods,the anther callus and roots,stems,embryoids gen-erated from the callus of Asparagus officinalis were studied.The callus could be divided intothree sorts,and there were obvious differences in morphological and structural characteristicsamong them.The embryoids generated asynchronously either on the surface of the callus orinside it.and their generating process and structure were similar to embryos.There were afew embryoids which could develop into plantlets.The stems produced on the surface of cal-lus had typical structure of tunica and corpus.The tunica contained only one layer of ceils.The morphological and structural characteristics of the roots and stems in vitro culture wereobviously different from that in the state of nature.
