摘    要:采用组织芯片技术研究KIF18A蛋白与卵巢癌发生的关系,并探讨该蛋白分子治疗临床卵巢癌的潜在价值.对比100例卵巢癌病患的癌组织与正常组织芯片,KIF18A蛋白分子在正常卵巢组织、卵巢癌、转移淋巴结中皆有不同程度的表达.在卵巢癌组织中的表达明显高于正常组织(p≤0.05).卵巢浆液性乳头状腺癌的不同分期中,Ⅰ期与Ⅱ期相比,在Ⅱ期中KIF18A蛋白分子的表达明显上调(p≤0.05).同时,淋巴结转移性浆液性乳头状腺癌中的KIF18A蛋白表达量高于非转移性的浆液性乳头状腺癌(p≤0.05).这些结果表明,KIF18A蛋白分子的表达强度与卵巢癌的肿瘤临床分期以及淋巴结转移有关,可作为卵巢癌检测的新型标志物.

Abstract:To investigate the function of KIF18A in the development of ovarian cancer and the potential values for treat ment of clinical ovarian cancer using the protein,tissue array analysis was used to detect relative quantity of KIF18A be tween different tissues.Compared normal or cancer tissue samples from 100 ovarian cancer patients,the correlation of KIF18A expression and clinico-pathological characteristics of serious ovarian tumors was analyzed.The expression of KIF18A was observed in normal ovarian tissues,ovarian cancer tissues and metastatic lymph nodes in different degrees.The expression of KIF18A in borderline serous cystadenoma and serious cystadenocarcinoma was significantly higher than that in normal ovarian tissues.Relative quantity of KIF18A in ovarian cancer tissues was higher than that in normal tissues(p≤0.05).Compared stage Ⅰ and Ⅱ of papillary serous adenocarcinoma,expression of KIF18A protein was significantly up-regulated(p≤0.05)in stage Ⅱ.Meanwhile,KIF18A expression in lymph nodes metastatic papillary serous adenocarcinoma was much higher than those in non-metastatic(p≤0.05).It was concluded that KIF18A was tightly associated to the staging of ovarian cancer and lymph node metastasis,which can be served as a novel target for the diagnosis and therapy for ovarian cancer.
