腾冲嗜热厌氧菌的超氧化物歧化酶的进化踪迹分析

摘    要:通过同源模建和分子动力学模拟,一个由腾冲嗜热厌氧菌的sod基因编码的蛋白质的可靠的分子结构被建立.对建立的结构进行进化踪迹分析识别出了11个踪迹残基.在它们当中,残基Asn36,Gly101和Glu158的位置遍布整个结构,无规可循;其余的残基都明显地聚于一个靠近铁原子的子类当中.这些结果表明,该基因编码含铁的超氧化物歧化酶;同时,那些铁原子附近的踪迹残基跟铁的绑定和催化功能直接相关.

Abstract:With homology modeling techniques and molecular dynamics simulations,a reliable molecular structure of the protein encoded by the gene sod from Thermoanaerobacter tengcongensis was built.Evolutionary trace analysis on this modeled structure reveal 11 trace residues.Among them,the residues of Asn36,Gly101and Glu158 are randomly scattered throughout the mapped structure;the other residues are all distinctly clustered in a subgroup near Fe atom. From these results,this gene encodes a Fe-depending superoxide dismutase.Furthermore,the trace residues around Fe atom are directly responsible for Fe binding and catalytic function.
