摘    要:建立了一种快速、简便、重现性好的测定中药制剂天参胶囊中小檗碱含量的非水毛细管电泳新方法,研究了乙酸氨浓度、离子强度、有机改性剂比例以及电压对分离行为的影响.实验结果表明,最佳缓冲液组成为40 mmol/L乙酸胺-2%乙酸-40%乙腈,最佳运行电压为25 kV,检测波长245 nm.分析物浓度与峰面积之间的回归方程为Y=-1664.05 787.16X,相关系数为0.9992.该方法已用于天参胶囊中的小檗碱含量测定,回收率为91.3%~94.6%.

Abstract:A simple, rapid, and reproducible nonaqueous capillary electrophoresis method has been developed for the separation and determination of berberine in a new kind of traditional Chinese medicine preparation. The effects of the concentrations of acetic acid and electrolyte, the ratio of organic solvent and the applied voltage on the separation were investigated. The optimum running buffer composed of 40 mmol/L ammonium acetate, 2% (V/V) acetic acid and 40% (V/V) acetonitrile in methanol. The applied voltage was 25 kV. The analyte was detected by UV at 245 nm. The linearity between peak areas and the concentrations of the analyte was also investigated and it exhibited excellent linear behavior over the concentration ranges (correlation coefficients was 0.9992). The method was successfully applied to determine berberine in Tianshen Capsule for the first time and the recovery of the constituent ranged between 91.3% to 94.6% for berberine.
