摘    要:利用1.8μm颗粒的色谱柱建立了HPLC法同时测定独活中六种香豆素和一种倍半萜衍生物含量的分析方法.研究结果表明,七种有效成分在线性范围内线性关系良好,回收率在95.8-101.9%范围之内,最低检出限和定量限分别低于0.06和0.14μg/mL;化学计量学的方法(相似度分析和聚类分析)可用于评价不同来源的独活的质量.本项研究验证了多组分含量测定和化学计量学方法联用可用于独活药材的质量控制,为独活药材的质量评价提供了研究手段.

Abstract:A simple and sensitive method has been developed for the simultaneous determination of six coumarins and one sesquiterpene derivative in radix angelicae pubescentis using HPLC-UV with a short column packed with 1.8 μm particles. The seven major constituents showed good regression(R > 0. 9987)within test ranges and the recoveries were in the range of 95.8- 101.9%. The limit of detections and quantifications for seven major constituents were less than 0.06 and 0. 14 μg/mL,respectively. In order to evaluate the quality of radix angelicae pubescentis from different areas,the chemometrics methods,including similarityanalysis (SA), hierarchical clustering analysis (HCA), were used to classify radix angelicae pubescentis samples according to their cultivated sources. The results showed that seven marker constituents could be applied to accurate discrimination and quality control for radix angelicae pubescentis by simultaneous determination. It was concluded that the combination of multicomponent analysis and chemometrics methods could be applied to quality control of radix angelicae pubescentis samples.
