摘    要:目的:通过mRNA结构预测软件预测出BCR—ABLmRNA二级结构,未形成二级结构的区域作为靶序列。方法:针对BCR—ABLmRNA二级结构中的单链区域设计四对siRNA,通过Lipotap脂质体作为转染载体去转染K562细胞。结果:所设计的四对siRNA其中有一对siRNA转染K562细胞后,K562细胞出现凋亡现象。结论:通过该方法有针对性的设计siRNA,避免了把mRNA的二级结构区域作为靶序列,提高了所设计siRNA的效果。

Abstract:Purpose : to predict BCR - ABLmRNA second - level structure through mRNA structure prediction software and with the area which has not formed second - level structure as target sequence. Method : according to the single chain area in BCR - ABLmRNA sec- ond - level structure, designing four pairs of siRNA and transferring IC562 cells with Liptotap liposome as transfer vector. Result: among four pairs of siRNA, only one pair caused the apoptosis of K562 cells after transferring them. Conclusion: by designing siRNA, avoiding taking the second - level structure of mRNA as the target sequence and improving the effect of sinRNA.
