Abstract:The aminoacylase(EC.3.5.1.14) was isolated and purified from Asperigillus oryzae No.3042 by the combination of ammonium sulfate fractionation and aqueous two phase system. The pure enzyme was immobilized on DEAE Sephadex A 25 to prepare immobilized enzyme, which was used on the optical resolution of D,L Alanine. In order to search better resolution condition, the enzymological properties of the native aminoacylase from Asperigillus oryzae 3042 and the immobilized aminoacylase were studied in this paper.
