| 摘 要： | 用江浙蝮蛇毒中纯化的中性磷脂酶Ａ２（ＮＰＬＡ２）免疫家兔，获得了多克隆抗体，抗体与抗原结合后，抑制了该抗原的神经毒性，而对其催化活力无影响，表明ＮＰＬＡ２有两个活性结构域，即神经毒性与催化活性分别位于两个活性位点上，利用免疫亲和层析纯化了ＮＰＬＡ２的多抗，经ＥＬＩＳＡ检测，与酸性ＰＬＡ２和碱性ＰＬＡ２均有免疫交叉反应，这提示了ＮＰＬＡ２与酸性ＰＬＡ２及碱性ＰＬＡ２的抗原决定簇有同源性。

| Abstract: | The polyclonal antibody was raised by pure NPLA2 from the venom of Agkistrodon halys Pallas in rabbit. The combination of antibody with antigen inhibited the neurotoxity of NPLA2, but it has no effect on catalytic activity. This fact shows that NPLA2 has two active doains. Immunoaffinity chromatography was used to purify anti-NPLA2. The results of ELISA told us that antibody of NPLA2 cross-reacted strongly with BPLA2 and APLA2. This indicated that some antigen determinants of NPLA2, BPLA2 and APLA2 are homologous. |
