通过紫外线诱变筛选溶磷细菌突变体

摘    要:为了选育具有优良溶磷性状的溶磷细菌突变体用于磷细菌肥料的生产,使用紫外线作为诱变剂,对溶磷细菌Y2041进行诱变处理.通过平板初筛和摇瓶培养复筛,得到溶磷能力明显提高的突变体Y2041-01,该菌株溶磷圈直径(12.02 mm)与菌落直径(2.48 mm)比值达4.85,比出发菌株Y2041的平板溶磷能力(2.22 mm)提高118.47%.将Y2041-01在含有Ca3(PO4)2的无机磷培养液中培养5 d,有效磷含量达33.85 mg/L,比出发菌株Y2041(28.73 mg/L)的溶磷效果提高17.82%.通过传代培养,磷细菌突变体Y2041-01的溶磷能力可以稳定遗传.

Abstract:In order to obtain the better mutants of phosphate-solubilizing bacteria,phosphate-solubilizing bacterium Y2041 was treated with UV.The mutant Y2041-01 with high ability was screened out by using the methods of transparent zone and shaking culture.The ratio of diameter of transparent zone(12.02 mm)to that of colony(2.48 mm)was 4.85,and the phosphate-solubilizing ability on plates of this mutant was increased by 118.47% compared with that of the original strain.When the mutant Y2041-01 was cultured for 5 days in the liquid medium containing Ca3(PO4)2,the available phosphate content got to 33.85 mg/L.Compared with that of the original strain(28.73mg/L),the phosphate-solubilizing ability of the mutant was increased by 17.82%.Furthermore,the subculture experiments indicated that the hereditary character of phosphate-solubilizing ability of the mutant Y2041-01 was stable.
