黄瓜离体雌核发育的过程及其早期生化变化研究

摘    要:本首次研究了黄瓜离体雌核发育的形态及细胞学过程,以及培养早期生理、生化的变化.结果表明:雌核发育在培养的前6d即已开始,24d时形成胚状体,40d时形成成熟的胚状体.雌核发育的前6d,外植体POD酶活性急剧上升,SOD先上升后下降,Q酶活性明显上升但低于非雌核发育,Ca^2 -ATP酶和Mg^2--ATP酶活性变化较小且均低于非雌核发育.雌核发育的外植体IAA和ABA含量明显下降,iPA,Zr,CA3和CA4含量均有所下降并明显低于非雌核发育外植体.

Abstract:Histological and early biochemical aspects of cucumber in vitro gynogenesis were studied. The results indicated that cucumber gynogenesis initiated within 6 days of culture. Embryoids were formed at the 24th day, mature embryoids were formed after the 40th day. In the first 6 days of culture, explant POD activity increased dramatically, SOD activity increased at the 3th day and decreased at the 6th day, Q enzyme activity increased obviously but lower than that of non-gynogenetic explant, Ca 2+-ATPase and Mg 2+-ATPase activity changed little and were lower than that of non-gynogenetic explant. IAA and ABA content decreased dramatically and were lower than that of non-gynogenetic explant. Zr, iPA, GA4 and GA3 content changed little and were obviously lower than that of non-gynogenetic explant.
