摘    要:为了提高微生物絮凝剂的产量,增强其絮凝效果,降低培养成本,以酱油废水替代发酵培养基对WN-2菌进行了培养,通过单因素培养条件优化,考察了外加碳源、外加氮源、培养时间、pH值等因素对微生物絮凝剂产生菌絮凝率的影响。实验结果表明,以预处理过的酱油废水作为廉价替代培养基,高效絮凝菌株WN-2可以在此条件下产生微生物絮凝剂。且WN-2最佳絮凝条件为:外加氯化钙浓度为0.2 g/L、无需添加碳源和氮源、培养时间为36 h、pH为7.0、摇床转速为160 r/min,在最佳条件下,对高岭土悬浊液进行絮凝测定,其絮凝率达到93.6%。因此,利用酱油废水作为微生物絮凝剂的替代培养基是完全可行的。

Abstract:In order to increase the yield of bioflocculants, improve the flocculantion efficiency and re- duce the cultivation cost, the WN-2 strain was fed on soy sauce brewing wastewater replaced of fer- mentation culture medium. Effects of carbon source, nitrogen sources, incubation time, pH value and other factors on the microbial floceulant's activity were analyzed through single factor test for optimi- zation of cultivation conditions. Results showed that bioflocculants can be produced by the WN-2 strain using the pretreated wastewater, a cheap substituting culture medium as the medium. The opti- mal conditions was that the strain was cultured adding 0.2 g/L calcium chloride concentration for 36 h ,on pH 7.0 and 160r/min conditions ,without carbon and nitrogen sources. Under above optimal conditions,flocculation of Kaolin suspension were determined and the flocculation rate was 93.6%. It is concluded that flocculants was produced by the WN-2 strain using soy wastewater as the alterna- tive medium is feasible.
