摘    要:用邻苯二甲醛和N-乙酰-L半胱氨酸作为柱前手性衍生化试剂,使用常规反相高效液相色谱法拆分DL广氨基酸对映体,对于不能达到基线分离的色谱峰,应用化学计量学方法计算分析,从而达到同时定量测定多种氨基酸对映体的目的。

Abstract:A pre-column derivatization method for the separation of amino acid enantiomers using ortho-phthaldialdehyde (OPA) and N-acetyl-L-cysteine (NAC) as tagging reagent followed by normal reverse-phase High-Performance Liquid Chromatography (HPLC) has been developed. Chemometrics algorithm was used to resolve the overlapped chromatographic peaks. At the same time, least square regress was used to obtain the factors between the resolved curves and the real peaks. The quantitative determination of amino acid can be achieved from a single run.
