团状雪灵芝提取过程中二十八烷醇的红外光谱跟踪分析

摘    要:对团状雪灵芝依次进行了石油醚、乙酸乙酯及乙醇提取,对雪灵芝原药材、各提取物及最终提取残渣进行红外光谱采集分析追踪二十八烷醇在提取分离过程中的去向.通过与二十八烷醇对照品的红外光谱对比分析发现,仅石油醚及乙酸乙酯提取物中可能存在二十八烷醇,且前者含量明显更高.二十八烷醇的红外吸收特征在二阶导数红外光谱中得到放大,除石油醚提取物及乙酸乙酯提取物外,雪灵芝原药材的二阶导数谱中也能明显看到二十八烷醇在723 cm-1处的特征吸收峰,而其他样品在此位置附近几乎无此吸收.本研究结果表明雪灵芝中二十八烷醇在提取过程中主要进入了石油醚提取物.红外光谱法是一种方便、快速、有效的检测方法,对于植物化学分离过程具有指导意义.

Tracking analysis of octacosanol in the extraction of arenaria polytrichoides by fourier transform infrared spectroscopy

Abstract:Arenaria Polytrichoides( AP) was extracted with petroleum ether( PE),ethyl acetate( EtOAc) and ethanol( EtOH) respectively. IR spectra of AP,all its three extractions and the last residue were collected for the purpose of tracking the migration of octacosanol in the extraction process. At the same time,IR spectra of pure octacosanol isolated from AP was collected and used as the comparative spectra. Results have shown that the characteristic absorption at 723 cm- 1of octacosanol can only be found easily in the two IR spectra of PE and EtOAc extractions respectively,and the peak intensity of the former is much stronger. The aforesaid absorption characteristic was magnified in the secondary derivative infrared spectra. Furthermore,the absorption peak at 723 cm- 1can also be observed in the spectrum of the raw material of AP,which can not be observed in 2D IR spectra of others' method. It concludes that most of the octacosanol in AP migrated to the PE extraction. Further more,FT- IR analysis method is a convenient,quick and efficient detection method which could guide the isolation and separation of the chemical composition of plants such as octacosanol.
