摘    要:采用高效液相色谱法,色谱柱Kinetex OOD-C18(100 mm×4.6 mm,2.6 μm),梯度洗脱流动相为乙腈-0.4%磷酸溶液,流速1 mL·min-1,检测波长350 nm,测定了不同产地紫花地丁中秦皮乙素的含量并建立其特征图谱,结果表明,不同产地15批紫花地丁的主要成分秦皮乙素含量差异较大,其中3批含量较低;应用指纹图谱相似度评价系统找出6个共有峰,指认3个色谱峰;15个样品相似度都在0.94以上。本研究建立的方法稳定、数据可靠,可以作为紫花地丁品质鉴别、质量控制及资源综合利用的依据。

Abstract:The aesculetin content of Violae Herba is determined and its characteristic high-performance liquid chromatography (HPLC) chromatogram is established in this study.Aesculetin content was determined by HPLC using the Kinetex OOD-C18 column (100 mm × 4.6 mm, 2.6 μm) and acetonitrile–phosphoric acid (0.4%) solution as the mobile phase in gradient elution; the flow rate was set at 1 mL/min and detection wavelength at 350 nm. The results showed that aesculetin, the main chemical constituent of 15 batches of Violae Herba obtained from different habitats, content varied widely. Among all batches, 3 had relatively low aesculetin content. The Similarity Evaluation System for Chromatographic Fingerprinting revealed 6 common peaks and 3 chromatographic peaks, and the similarity of the 15 samples was above 0.94. The method established in this study can consistently produce reliable data and provide evidence for the quality determination and control of Violae Herba and wide application of resources.
