测菌体的OD值法探索松口蘑种子最佳生长周期

通过对松口蘑液体发酵工艺进行研究,实验通过测菌体OD值的方法初步得到松口蘑最适培养基:蔗糖30g/L、可溶性淀粉10g/L、酵母膏5g/L、KH2PO40.5 g/L.发酵条件:生长温度26.5℃、摇床转速:170 r/min、pH值:4.5、接种量:10%.首先通过传统的称量菌体干重的方法得到最佳生长周期是11-12 d,菌体得率是11.37 g/L,测菌体OD值探索菌体的生长周期,具有比一般传统方法例如测生物量的方法更加简便有效,并具有较高的准确度,为下一步进行菌根的研究打好基础。

Researching the Best Growth Cycle of Tricholoma Matsutake by OD

The best culture Medium of Tricholoma matsutake was determined by studying the liquid fermentation technique of Trieholoma matsutakc . Sucrose： 30g/L ,Soluble starch ：10 g/L , Yeast 5 g/L ,KH/PO4 0.5 g/L. The best fermenting condition is： 26. 522 , rotating speed 170 r/rain, pH 4.5, inoculating amount 10%, 100mL culture medium per 250 mL flask. Under these conditions, the mycelia reach 11.37 g/L weight of Dry cell after 11 -12 d＇s fermentation. In addition the OD of Tricholoma matsutake is determined in this Process, which is a much more efficient way.
