凤尾菇线粒体DNA片段的克隆与同源性分析

将Ｓａｕ３ＡＩ部分酶切的凤尾菇线粒体ＤＮＡ插入质粒载体的ｐＢＳ＾＋的ＢａｍＨＩ位点，然后以凤尾基线粒体ＤＮＡ为探针与重组质粒ＤＮＡ进行斑点杂交，筛选到６个阳性克隆，选取其中２个阳性克隆的插入片段分别与来６个属的１３个食用菌品种的线粒体ＤＮＡ进行Ｓｏｕｔｈｅｒｎ杂交，其结果显示两探针能与所有测定的侧耳属品种的线粒体ＤＮＡ杂交，并具多态性，但与其它属线粒体ＤＮＡ只有不同程度的杂交，两探针中Ｈ４的杂交范

CLONING AND HOMOLOGOUS ANALYSIS OF MITOCHONDRIAL DNA FRAGMENTS FROM PLEUROTUS SAJOR CAJU

Sau 3AI partially digested mitochondrial DNA fragments of pleurotus sajor caju were cloned into Bam HI site of pBS .Six positive clones were obtained by using labeled mtDNA of P.sajor caju as probe hybridized with recombinant plasmid DNA.Two positive clones were chosen for the isolation of inserted fragments which were used to study their homology with other mtDNA of thirteen cultivated mushroom strains belonged to six genera.The results showed that both of the cloned mtDNA fragments,assigned H2 and H4,could hybridize with mtDNA of all eight tested strains from Pleurotus and displayed a polymorphism of hybridized band pattern.But,when they were used to hybridize with the mtDNA from other genera,there was a difference between these two fragments.H4 showed a broad hybridization pattern,H2,however,could only hybridize with the mtDNA from a few strains.
