硝态氮和氨态氮对玉米幼胚愈伤组织的影响

摘    要:以自交系87-1和组合87-1×5237、A188×87-1的幼胚为材料,在降低氮含量的筛选培养基(8个浓度N0,N25,N50,N75,T0,T25,T50,T75)上筛选三次,进行生长量的测定、筛选后培养基pH值的测定和愈伤组织类型变化情况的观察,结果表明:降低培养基中氮含量,愈伤组织生长变慢,Ⅱ型愈伤组织逐渐减少,Ⅲ型和混合型增多;硝态氮系列浓度筛选培养基的pH值下降幅度较大;硝态氮对玉米愈伤组织的影响大于氨态氮.

Abstract:Using one inbred line(87-1) and two hybrids(87-1×5237?A188×87-1) embryo of maize as the explants,callus was selected on eight kinds of medium which had different concentrates of nitrogen(N0,N25,N50,N75,T0,T25,T50,T75) three times.The growth amount of callus and the value of pH were determined and the type of callus was observed.The results indicated that the growth amount and quality of callus descended with the concentrates of nitrogen reducing.The value of pH was also varied after selecting.With nitrogen concentrate decreasing,the type Ⅱcallus decreased,type Ⅲand combined type increased.The range of the value of pH on nitrate selecting medium varied largely,nitrate had greater influence on callus of maize than ammonia.
