青檀基因组DNA提取方法的优化

摘    要:以青檀(Pteroceltis tatarinowii Maxim.)的叶片为材料,采用改良高盐低pH值法、改良SDS法、改良CTAB法和试剂盒法4种不同方法提取其基因组DNA,并进行电泳分析、质量检测及ISSR引物扩增检测.结果表明,改良CTAB法优于其它方法,提取的DNA质量较好,纯度较高,能满足进一步的实验要求.

Abstract:The genomic DNA was isolated from the leaf of Pteroceltis tatarinowii Maxim.by using the improved high salt and low pH method,improved SDS method,improved CTAB method and genomic DNA kit.The quality of the DNA was examined by agarose gel electrophoresis,spectrophotometry and PCR amplification of ISSR.The result showed that the method of improved CTAB was obviously better than the others.The DNA sample obtained by the improved CTAB method was pure and had good quality which could satisfy the need for further...
