纽荷尔橙内皮中SOD的纯化及部分性质研究

摘    要:利用有机溶剂分级沉淀法从纽荷尔橙皮内层中分离出超氧化物歧化酶(SOD),研究了分离纯化的最佳工艺条件,并对其性质进行了探讨.结果表明,当纽荷尔橙内皮(g)与磷酸缓冲液(mL)之比为13,磷酸缓冲液pH值为7.8,提取时间为20min,氯仿-乙醇混合物量为提取液的0.25倍,丙酮量为粗酶液的1.5倍时,分离效果最佳.该SOD酶型是Mn-SOD,30℃-60℃,pH值4-8,活力稳定.研究结果为SOD提供了新酶源.

Abstract:The organic solvent graduation precipitation method was used to separate SOD from inner layer of newhall orange.The result indicated:when the inner layer(g) of newhall orangeand phosphoric acid buffer solution(ml)ratio was 1:3,and phosphoric acid buffer solution's pH was 7.8,20min's extraction times,and when chloroform-ethyl alcohol mixture was 0.25 times of the extracting,and cetone is 1.5 times of the thick enzyme fluid,the separation result was better than any other conditions.This study supplied new enz...
