摘    要:端粒酶是一种核蛋白,在真核生物体内,通过将端粒重复顺序转移到染色体末端而维持染色体的稳定.端粒酶活性存在于生殖细胞和永生癌细胞,但在正常细胞中却极低乃至无法检测.我们使用EB染色简化了端粒酶活性检测的传统的TRAP(Telom eric Repeat Am plification Protocol)法.结果表明,对粗抽提物而言,EB染色具有足够的可信度和灵敏度,同时又经济省时,便于推广使用

Abstract:Telomerase is a ribonucleoprotein enzyme adding telomeric repeats onto the ends of eukaryotic chromosomes to maintain telomeres that are essential for chromosomal stability. Telomerase activity has been detected in germline cells and immortalized cancer cells, but in normal cells is either low or undetectable. The Telomeric Repeat Amplification Protocol(TRAP) assay had been modified to detect telomerase activity qualitatively by using ethidium bromide(EB). The present assay is sensitive, reliable and time saving for the detection of telomerase activity in crude extract.
