浓香型大曲微生物群落结构的PCR-SSCP分析条件优化

摘    要:文章研究了浓香型大曲微生物群落结构的PCR-SSCP分析条件的优化,结果表明:PCR产物100℃变性5 min后,在交联度为49:1,浓度为8%的聚丙烯酰胺凝胶中,无甘油,4℃条件下200 V电泳15 h,可得到较理想的图谱。

Abstract:The optimal conditions of PCR-SSCP Technology for the analysis of microbial communities in Luzhou-flavor Daqu is studied in the paper.The results show that under the condition of the concentration of the gel being 8%,the proportion of acrylamide and N,N'-methylene-bisacrylamidei being 49:1,without glycerine,denaturing at 100℃for 5 min,4℃, 200 V,electrophoresis 15 h,perfect spectrogram can be achieved.
