高产碱性蛋白酶工程菌的构建及鉴定

摘    要:利用原生质体融合技术,使碱性蛋白酶生产菌2709与枯草杆菌BD105(含有碱性蛋白酶基因克隆载体pDW2)杂交,得到1株高产碱性蛋白酶的工程菌A16.A16的表型与生长特征与2709相似,相同条件下发酵酶活性比2709菌株高50%,摇瓶发酵的产酶水平最高可达30000u/mL。用菌落的原位杂交鉴定出该菌株携有双亲的遗传物质.

Abstract:A engineering strain with a high ability of producing alkaline protease was obtained by fusing protoplasts of Bacillus licheniformis 2709 and Bacillus subtilis BD 105 [pDW 2(pMK 4 and alkaline protease gene)]. Cell proliferation aspect and phenotype of A16 was similar to that of 2709, and its activities of fermentation were 50%, higher that that of 2709 under the same conditions. The maximum activities of protease were 30000 u/mL from A16 in the optimum fermenntation. In situ hybridization data indicated that A16 carried genes from BD105 (pDW 2) and B. licheniformis 2709.
