禽类骨髓细胞染色体标本制备实验方法的改进

以鸡和鹌鹑为实验材料,介绍一种改进的禽染色体标本制备技术。通过选取适宜的实验动物,改进秋水仙素的剂量、低渗的温度和时间、离心速度等关键步骤,阐述了有关染色体标本制备的实验方法和条件,建立了一套实用性强、染色体标本清晰、结果稳定、成本较低、简单易行的禽类染色体制片技术,为农业院校本科生、研究生进一步研究染色体核型和带型提供实验方法。

An Improved Experiment Method for Preparing the Chromosomes Sample of the Poultry's Marrow-cell

The chicken and the quail were used as experiment materials.The paper introduced the poultry’s chromosome samples technology which has been improved.The study explained experimental method and condition of the poultry’s chromosome samples by choose suitable experiment animals,improved the key steps of colchicine dosages,temperature and time on low-osmolar,centrifugal speeds.It was established to the poultry’s chromosome samples technology with strong practicality,clearer chromosome samples,stability result,lower cost,simple and feasible method.This will provide an experiment method to study on the karyotype and banding of chromosome for the undergraduate and postgraduate students of agriculture university.
