海带硫酸化多糖的提取及纯化

摘    要:对海带硫酸化多糖的水提法与酶解法进行比较,发现酶提法效果较好,粗提得率较水提法提高了85.9%,纯化后得率提高了43.6%.纯化过程中,通过正交实验得出CTAB沉淀LPS的最佳最佳优化条件为1%粗糖液:5%CTAB(V/V)为3:2,沉淀时间6h,离心时间12min.经单因素实验确定了树脂D315的脱色条件为:1%多糖液与树脂的体积质量比为为100:1(V/W),脱色时间6h,pH值5.0,温度40℃.

Extraction and Purification of La Minaria Polysaccharide Sulfate

Abstract:The polysaccharide sulfate was extracted from La minaria japonica by water extraction and enzyme extraction. The result showed that enzyme extracting method was more effective than that of water extraction,the extracting rate of crude LPS and pure LPS was increased by 85.9 0% and 43.6 % respetively,compared with that of water extraction. The crude LPS was precipitated using CTAB as precipitant. Through orthogonal experiments,we obtained the optimal technological parameter, ratio of 1 LPS and 5% CTAB 3 : 2, precipitation time 6h, centrifugal time 12 min. Decolorizing experiments were carried out by single experiment. The best conditions for deeoloration were deter mined as follows:D315 resin,ratio of 1% LPS and resin was 100: 1(V/W) ,Time=6 h,pH=5.0,T=40 ℃.
