功能性消化不良重叠肠易激综合征动物模型的建立

目的探讨建立功能性消化不良(Functional Dyspepsia,FD)重叠肠易激综合征(Irritable Bowel Syndrome,IBS)动物模型的方法。方法将大鼠随机分为3组:空白对照组(A),先夹尾刺激后灌胃组(B),先灌胃后夹尾刺激组(C),利用行为分析仪对大鼠旷场实验进行分析,了解大鼠的焦虑、抑郁程度;同时,取大鼠的胃窦进行组织学观察。结果 B组的变化比C组更加接近于人类的发病情况。与A组相比,模型组大鼠旷场实验有统计学意义(P<0.05)。两模型组之间除一般状态和旷场实验以外也存在显著性差异(P<0.01)。结论大黄泻下法结合夹尾应激法能成功建立FD-IBS重叠征模型,夹尾刺激和灌胃按不同顺序实施均能成功建立,且先夹尾刺激后灌胃组建立的重叠征模型优于先灌胃后夹尾刺激组。

The Establishment of Model of Functional Dyspepsia Overlapping with Irritable Bowel Syndrome in the Rats

Objective To establish the rat models of functional dyspepsia overlapping with irritable bowel syndrome.Method 30 Wistar rats were randomly divided into the control group （A group）,the tail clamp stimulation first and then gavage group （B group）,and the gavage first and then the tail clamp stimulation group （C group）.The open field test was performed to evaluate the degree of anxiety and depression,and the histological changes of the gastric antrum were observed.Result Compared with C group,the changes were more close to the incidence of human in B group.The differences in indexes of open field test before and after developing of the model were significant （P ＜ 0.05） in B and C group,and there was a significant difference in sucrose consumption between B group and C group.Conclusion Rhubarb purgation before tail clamp or tail clamp before rhubarb purgation could develop the model of functional dyspepsia overlapping with irritable bowel syndrome in the rat,and the model developed by the tail clamp stimulation first and then gavage method is superior in some respects to that developed by gavage first and then the tail clamp stimulation method.
