抗人尿激酶单克隆抗体的制备

摘    要:采用常规免疫和脾内免疫相结合的方法,利用杂交瘤技术得到两个能分泌抗人尿激酶单克隆抗体的杂交瘤细胞株10H10,10D12。它们分泌的单克隆抗体主要识别Mr.54,000的高分子量尿激酶和Mr.18,000的尿激酶A链。两种单克隆抗体与胰蛋白酶,胰蛋白酶原,胰凝乳蛋白酶和胰凝乳蛋白酶原等丝氨酸蛋白酶和酶原无交叉反应。

Abstract:Combining ordinary immunization with intrasplenic immunization, tow hybridoma lines 10H10 and 10D12 have been screended out, which secreted monoclonal antibodies against human urokinase by fusion between SP2/0 myeloma cells and spleen cells from a BALB/c mouse immunized with urokinase, monoclonal antibodies 10H10 and 10D12 strongly bound to urokinase and showed no crossreaction with trypsin, trypsinogen, chymotrypsin and chymotrypsinogen. Immunoblotting of urokinase showed that McAbs 10H10 and 10D12 recognized either the 54,000 dalton urokinase or the 18,000 dalton urokinase chain.
