微囊藻毒素对Hela细胞和Vero细胞的毒性效应

摘    要:MCYST-LR,MCYST-RR,MCYST-YR稀释后的毒素溶液处理Hela细胞和Vero细胞后得到如下结果:1.3种毒素溶液对培养的Hela细胞均有明显的抑制作用和毒性作用,其毒性作用与浓度成正比,致死细胞边缘不整齐不规则,细胞呈脱水状态。倒置显微镜下可见细胞变黑无折光性。高浓度毒素溶液(包括MCYST-LR,RR和YR3个样品)作用细胞24h后,细胞完全致死脱落,3个样品间无明显差别;2.

Abstract:Microcystins are isolated and purified by HPLC from the blue green algal (cyanobacterial) Microcysts bollom collected in Dianchi Lake of the Southwestern China. The Microcystin LR(MCYST LR), YR(MCYST YR) and RR(MCYST RR) which are diluted into three similar concentrations in advance are added to the cultured Hela and Vero tumor cells. The results after incubating for 24 hrs show: 1.all three microcystins represent toxic and inhibitory effect on the growth of the two tumor cells, furthermore, the intensity of the effect is in correspondence with the concentration of the toxins. It is almost the same that the two tumor cells are caused death after 24 hrs with higher concentrations of the three microcystins. The dead tumor cells are irregular and dry. 2.the Hela cells seem to be more poisobned than the Vero cells in the same concentrations of the toxin liquids. 3.Microcystin YR represents more inhibitory effect on the Hela cells than Microcystin RR does. 4.under higher toxin concentration, the observation over long term on the test is ineffective for the shortage of the nutrition and that may be due to the relatively low content of the original microcystins.
