n-3PUFA高脂饮食导致SD大鼠胰岛素抵抗

摘    要:目的分析长期饲喂n-3多不饱和脂肪酸(PUFA)高脂饮食对大鼠胰岛素敏感性的影响。方法雄性SD大鼠随机分为2组,对照组(NC)给予普通日粮,高脂组(TUF)给予富含n-3PUFA的高脂日粮。喂养16周,记录采食量及体质量,用高胰岛素—正常血糖钳夹技术评价大鼠胰岛素敏感性。结果 TUF组体质量与NC组没有统计学差异(P〉0.05),基础血糖及胰岛素与对照组没有统计学差异(P〉0.05),葡萄糖输注率(GIR)显著低于NC组(P=0.031)。结论长期过量摄入n-3PUFA导致大鼠胰岛素抵抗。

Abstract:The n-3 Polyunsaturated fatty acids(PUFA) are increasingly consumed as food supplements;however,the side effects of it,especially at high doses,remain unclear.The aim of this study was to analyze insulin sensitivity in SD rats fed with high-fat diet rich in n-3 PUFA.Normal male SD rats were randomly divided into two groups,including normal control group(NC) and high n-3PUFA formula diet group(TUF).Rats were fed for 16 weeks,diet intake and weight were recorded,insulin sensitivity was estimated by euglycemic-hyperinsulinemic clamp.By the end of the experiment,weight was similar in the two groups,fasting plasma glucose and plasma insulin in TUF group were not significantly different compared to NC group(P>0.05),GRI in TUF was obviously lower than that in NC group(P=0.031).In conclusion,Overdosage intake of n-3PUFA significantly decreased insulin sensitivity in rat.
