4-溴苯并[b]噻吩的合成及表征

摘    要:利用3-溴苯硫酚和2-溴-1,1-二-甲氧基乙烷为起始原料,经过SN2亲核取代和分子内环化两步反应合成了4-溴苯并[b]噻吩.通过对溶剂、温度、反应时间、多聚磷酸(PPA)用量等影响反应收率的主要因素的考察,得到了合成目标化合物的较佳反应条件.第一步较优条件:选择DMF作为溶剂,反应温度35~45℃;第二步较优条件:100mL氯苯作溶剂情况下,PPA投料量20 g,反应温度110℃.实验结果表明,该反应条件温和,后处理简单,并且目标产物的收率(64.2%)令人满意.产物的结构通过IR,GC-MS和1HNMR进行了确证.

Abstract:4-Bromobenzo[b]thiophene was synthesized using 3-bromothiophenol and 2- bromo-l,l-dimethoxyethane as starting materials by SN2 nucleophilic substitution and intramolecular cyclization. The optimum conditions were obtained by examining the effects of solvent, temperature, reaction time and PPA dosage on product yield. The obtained optimum conditions of the first step were DMF as solvent and resction temper- ature 35--45 ℃. In the second step, 100 mL of chlorobenzene as solvent, PPA 20 g and reactiom temperature 110℃. The results showed that the reaction conditions were mod- erate and the yield of product was 64.2 % with simple workup. The structure of product was characterized by IR, GC-MS and 1H NMR.
