摘    要:试验以成年态纽荷尔脐橙茎段为外植体,诱导产生不定芽改进微芽嫁接成活率与脱毒率.结果表明:半木质化茎段作为培养外植体污染率最低,不定芽诱导的最佳培养基为MT++6-BA1.0mg/L+NAA0.2mg/L+GA30.2mg/L.将诱导的嫩芽进行微芽嫁接,成活率超过80.8%,对照成活率为61.3%,聚合酶链式反应(PCR)检测脱毒率达100%.

Abstract:Adventitious buds were induced with mature stem segments of Newhall navel orangeCitrus sinensis(L_*)Osbeck]as explants in an in vitro culture experiment to improve the survival rate and virus elimination rate in shoot-tip micro-grafting.Contamination rate was the lowest when half lignified stem segments were used as explants.MT++ 6-BA 1.0 mg/L+NAA 0.2 mg/L+GA_3 0.2 mg/L was the best medium for adventitious bud induction.More than 80.8% of the regenerated shoots survived when used in shoot-tip micro-grafting,in contrast to 61.3% for the control.PCR test showed that 100% of the grafted plants were virus free.
