| 摘 要： | 双向凝胶电泳技术是蛋白质组学研究的基础性技术平台 ,如何得到一张高质量的双向凝胶电泳图谱是进一步深化研究的关键 .该文利用人永生化食管上皮细胞系SHEE为材料 ,采用IPG DALT系统双向凝胶电泳技术 ,通过对样品的制备及预处理、第一向等电聚焦、平衡和两向间的转移等关键步骤进行改进和优化 ,获得了高分辨率、高重复性的双向凝胶电泳图谱 |

| Abstract: | Two-dimensional gel electrophoresis(2-DE)is one of the key techniques in proteome research and how to obtain a high-quality 2-DE map is a matter of great importance.In this paper is reported a optimization scheme for IPG-DALT two-dimensional gel electrophoresis which.using the immortalized human fetal esophageal epithelial cell line SHEE as the material,and optimizing such key steps as sample preparation and pre-treatment.isoelectric focusing,equilibrium and transfer,etc.,obtained a 2-DE pattern with high resolution and good reproducibility. |
