培养肝癌细胞有丝分裂中的动态

<正> 一、实验目的:观察肝癌细胞有丝分裂中的细胞运动和归转并探讨其机制和生物学意义。二、材料与方法:使用 BEL—7402肝癌细胞系培养于含20%小牛血清的199培养基中。原瓶传代后48小时,在倒装显微镜直视下按水平方向摇瓶,得到基本上同步于 M 期的肝癌细胞,转接到特制的双盖片培养瓶内(另有短文介绍这种培养瓶),置 Olympus(IMT型)倒置显微镜下保温35℃作追踪观察,并作不定时摄影记录。

SWIMMING MOVEMENT PERIOD OF MITOTIC HEPATOMA CELLS

A hepatoma cell line,BEL—7402,incubated at 35C in M199 and 20% calf serum to study mitotic cells.A rotatory swimming movemnt period in the mitotic phase of the cell cycle was noted,it took 5 up to 110 minutes.while the cells stoped rotation,they began to anchore and devide,some of which showed very complicated morphological changes(Plate),and finaaly,they spread out in 240—400 minutes since the departure off the original spreading places. Several problems are raised and discussed briefly concerning the mechanism of rotatory swimming movement,the possible role in metastasis,the biological significance of morphological changes.
