| 摘 要： | 以不同西伯利亚白刺组织为材料 ,利用TRIZOL试剂快速提取法、异硫氰酸胍法、苯酚法和CTAB法提取白刺总RNA ,通过RNA的产率、纯度、电泳图谱等分析来确立适用于白刺总RNA分离的方法 研究结果表明 ,CTAB法提取的RNA具有 2 8SrRNA、 18SrRNA和 5SrRNA三条较清晰的条带 ,很少有降解 ,其A2 6 0 /A2 80为 1.893～ 1.90 1,A2 6 0 /A2 30为 1.80 7～ 2 .4 87 具有较高的纯度 其他三种方法获得的RNA纯度较低 ,降解严重 ,有较多小分子和盐存在 ,RNA无明显条带出现 ,而是以小分子RNA弥撒状分布 将CTAB法提取的RNA逆转录成cDNA ,使用随机引物进行PCR扩增 ,PCR条带清晰 ,分布均匀 ,背景小 该结果进一步证明了CTAB法提取的RNA具有很高的质量与纯度 ,可以满足下一步的研究

| Abstract: | Our work is to establish a suitable method of RNA isolation from Nitraria L. through analysis the rate of yield, purity of total RNA, which obtained by the methods of TRIZOL regent, Guanidine Thiocyanate, Phenol, and CTAB. The results demonstrate that the RNA isolated by CTAB showing three clear bands of 28S rRNA, 18S rRNA and 5S rRNA respectively, and its purity is higher than that of the other methods though its yield is lower slightly. The ratio of A260/A280 is 1.893 ～1.901 and the value of A260/A230 is 1.807 ～2.487. RNA isolated by other methods degrades seriously and disperse. The electrophoresis figure by RT-PCR show that there are ideal amplified bands using the RNA templet isolated by the method of CTAB reagent and random primer. |
