摘    要:目的建立鹿鞭及其伪品随机扩增DNA多态性(RAPD)鉴定方法,为鹿鞭的鉴别提供可靠依据.方法采用柱层析法从鹿鞭及其伪品中提取线粒体DNA并进行随机DNA多态性扩增.结果正品鹿鞭(梅花鹿鞭和马鹿鞭)与其常见伪品(牛鞭)线粒体DNA随机扩增产物的电泳图谱有明显区别,条带数量和位置均不同,说明本方法可以有效区分正品鹿鞭与其常见伪品.结论 RAPD技术可为鹿鞭及其伪品的鉴别提供一种简单、有效、可信度高的方法.

On Application of RAPD Technology on Penis Cervi Identification Based on mt DNA
Abstract:Objective To establish the identification method of Penis Cervi from its adulterants by RAPD technology,and to provide a reliable evidence for Penis Cervi identification. Method Column chromatography method was used to extract and purify mt DNAs from Penis Cervi and its adulterants,and the DNA fragments were amplified by RAPD technology. Results The results showed that there was a great difference in the number and brightness of the stripes of RAPD products between Penis Cervi ( Gervus nippon Temminck whip and Cervus elaphus Linnaeus whip) and its adulterants(Bull whip). Conclusion The technology of RAPD could be a simple,effective and reliable method for the identification of Penis Cervi from its adulterants.
