已烯雌酚对蝌蚪抗敌敌畏诱导氧化损伤的作用研究

摘    要:研究已烯雌酚(DES)对中华大蟾蜍(Bufobufo gargarizans)蝌蚪抗敌敌畏(0,0-二甲基-0-(2,2-二氯乙烯基)磷酸酯,DDVP)诱导的氧化损伤的作用.方法:蝌蚪(30~35期)随机分为5组:I(0mg/LDDVP+0mg/LDES)为空白组;Ⅱ(12.9mg/LDDVP+0mg/LDES)为单独DDVP处理组;Ⅲ(12.9mg/LDDVP+1.0mg/LDES);1V(12.9mg/LDDVP+2.0mg/LDES);V(12.9mg/LDDVP+3.0mg/LDES)为DDVP和DES联合处理组.在染毒24h和72h时测定蝌蚪肝组织中超氧化物歧化酶(SoD)和谷胱甘肽过氧化物酶(GSH-Px)的活性以及丙二醛(MDA)的含量,计算抗氧化指数.其结果在DDVP(12.9mg/L)存在的条件下,随着DES(1.0mg/L)的加入和浓度的依次加大(2.0mg/L,3.0mg/L),SOD活性呈下降趋势,72h时具显著性(P〈0.05);GSH—Px活性呈先升高后降低的变化趋势,在24h和72h均具极显著性(P〉0.01);MDA含量呈先降低后升高的变化趋势,72h时具显著性(P〈0.05)。其中联合处理组Ⅲ的抗氧化能力最强.结论:DES对DDVP诱导的氧化损伤作用具有拮抗效应,能有效地增强中华大蟾蜍蝌蚪的抗氧化能力.

Abstract:The effect of diethylstilbestrol (DES) on antioxidation in tadpoles (Bufo bufo gargarizans) exposed to dichlorovos (0,0 Dimethyl-0-0 (2,2-Dichloro ethylene) Phosphate,DDVP) was studied. Methods:Tadpoles (30-35 tadpole stage) were randomly divided into 5 groups, as control group I (0 mg/L DDVP+0 mg/L DES), single DDVP group Ⅱ (12.9 mg/L DDVP+0 mg/L DES), experimental groups Ⅲ (12. 9 mg/L DDVP+I. 0 mg/L DES ), Ⅳ (12.9 mg/L DDVP+2.0 mg/L DES)and V(12.9 mg/L DDVP+3.0 mg/L DES). The activities of superoxide dismutase and glutathione peroxidase (GSH-Px) and the content of malondialdehyde (MDA) in liver of tadpoles were measured after 24 h and 72 h exposure, and the antioxidant index was computed. Results. With DDVP being 12.9 mg/L, the activity of SOD de- creased as DES was added and its concentration increased. After 72 h exposure, the activities of SOD reached the significant level(P〈0.05). The activities of GSH-Px increased at the first stage and then decreased, indicating the most significant difference(P〈0.01) at 24 h and 72 h exposure. The MDA accumulation which showed the adverse tendency as GSH-Px did, indicated the significant difference(P〈0.05)at 72 h exposure. The experimental group Ⅲ had the strongest antioxidation. Conclusion: DES has the strongest antagonistic effect on oxidative damages induced by DDVP, which can enhance antioxidation in tadpoles of Bu fo bu fo gargarizans effectively.
