摘    要:胎儿脐血作为富含CD34+细胞等造血干/祖细胞一个新来源,具有细胞增殖速度快,来源充足,收集安全等特点;脐血移植同样具有安全性好,效率高,费用低等优点。随着脐血干细胞移植临床应用的成功,欧美许多发达国家已经着手建立大规模脐血库以满足临床需求。建立脐血库技术上要求改善细胞冷冻保护剂组成,控制降温速率和细胞复苏方法,以获得理想的脐血干/祖细胞收率。同时由于脐血库液氮保存设备费用昂贵,需要预先分离脐血单个核细胞以减少样本冻存体积,以满足建库实际需求。

Abstract:Cord blood, as a new resource of hematopoeitic stem/progenitor cells, has a enrichment of CD34 + cells. The advantages of cord blood hematopoeitic stem cells are high speed proliferation, rich concentration and safety collection. Cord blood transplantation also has expected advantages such as safety, high efficiency and low expense. With the clinical success of cord blood transplantation, large scale cord blood bands are being built in many advanced countries in Europe and North America to meet the clinical application. The key technology of cord blood bank building for the ideal recovery of stem/progenitor cells includes improvement of cell cryopreservation prevention combination, cryopreservation rate control and cell recovery method. Cord blood separation and volume reduction is first needed to meet thd practical bank requirement for the high expense of liquid nitrogen preservation equipment.
