摘    要:通过收集污染区男性精液标本30份和30份对照地区成年男性的精液标本,提取精子RNA,标记后与miRCU-RYTM LNA Array芯片杂交,分析环境污染与对照地区成年男性精子中miRNA的表达情况。结果表明环境污染与对照地区男性精子共182个miRNA存在表达差异,在环境污染地区73个miRNAs显著上调,109个m...

Abstract:30 semen samples from electronic waste polluted environment and normal control environment site.Total RNA were harvested using TRIzol(Invitrogen) and RNeasy mini kit(QIAGEN) according to manufacturer’s instructions.Samples were labelled using the miRCURYTM Hy3TM/Hy5TM Power labelling kit and hybridized on the miRCURYTM LAN Array(v.14.0).The samples were hybridized on a hybridization station.There were 73 significantly up-regulated miRNA and 109 down-regulated miRNA in the expression profile of adult man sperms in electronic waste polluted environment compared with the miRNA expression profile of adult man sperms in normal control environment.Construction of miRNA profiles in adult man sperm of environmental polluted sites can help to analyze the molecular mechanism of the pathogenesis of spermatogenesis by environmental pollution.
