以光生物素标记DNA的点杂交比色法迅速鉴定类单胞菌的研究

摘    要:应用光生物素标记参考菌株DNA比色法以DNA-DNA点杂交对类单胞菌属的分离菌株及参考菌株进行遗传学鉴定。在强日光灯下,光生物素标记DNA15min。以少量菌体法提取被测菌株的DNA,并将此DNA点种在硝酸纤维膜上;碱性磷酸酶偶联链霉抗生物素基物显色法DNA-DNA点杂交,颜色图形分析仪测定点杂交颜色强度。该方法鉴定结果与数值分类法的结果和复性率定量测定DNA-DNA同源性的结果基本相符。光生物素标记DNA比色法DNA-DNA点杂交鉴定类单胞菌是一种快速而简便的方法。

Abstract:A rapid genetic identification of the isolated and reference strains of Alteromonas by colorimetry dot hybridization has been studied. Under sunlight, the ,DNA of reference strain was labeled with photo-biotin within 15 min. DNA obtained from a 50 ml culture of isolated strain could be sufficiently used for several hybridization experiments on a spotted nitrocellulose filter. Hybridized spots were visualized with alkaline-phospha-tase-conjugated streptavidin color-detection method. Hybridized filter was analyzed with a color graphic analyzer. The results of this study coincided with those of the numerical taxonomy and of the DNA homology of Alteromonas. This identification method for Alteromonas is rapid and simple.
