摘    要:目的:探讨人端粒酶催化亚单位hTRT基因在肝癌组织中的表迭情况.方法:用兔抗人端粒酶催化亚单位多克隆抗体作为一抗,生物素化羊抗兔IgG作为二抗,对肝癌组织石蜡切片采用免疫组织化学方法测定hTRT基因的表达.结果:30例肝癌组织中25例检出hTRT阳性(25/30),正常肝组织无l例阳性.结论:免疫组织化学法检测hTRT敏感性高、特异性强、重复性好,较其他方法简便、快速、经济且无放射性核素污染等缺点,临床具有推广价值.

Abstract:Objective To investigate human telomerase activity and clinical application by immunohistochemistry assay. Methods Rabbit anti-human telomerase catalytic subunit polyclonal antibody was used specific antibody and sheep anti-rabbit IgG was used the second antibody. Paraffin slice samples of tumor tissues were detected by immunohistochemistry assay. Results Telomerase activity of 25 cases was positive in 30 cases of liver cancer tissues. The positive rate of telomerase activity was 25/30. Conclusion Immunohistochemistry assay for telomerase activity is sensitive, reproducible, simple, rapid, cheap and without radioisotopes pollution and damage. Therefor, it can be applied extensively clinically.
