摘    要:以苔藓植物为材料,比较快速提取法、SDS法和改良CTAB法(自行设计)提取苔藓植物总DNA的效果.结果表明:改良CTAB法提取的基因组DNA质量最好(A260/A280为1.83~1.86;A260/A230为2.05~2.50),快速提取法次之(A260/A280为1.64~1.73;A260/A230为1.68~1.77),SDS法较差(A260/A280为1.51~1.61;A260/A230为1.56~1.76).改良CTAB法提取的苔藓植物的DNA适合于作为PCR的模板,并成功地进行了RAPD扩增.

Abstract:With Bryophytes used as ingredient,to compare total DNA extracted effects of the different three methods such as the fast extraction,SDS and the improved CTAB method which was designed by our research group.The results showed that the improved CTAB method had a remarkable superiority overothers(A260/A280 was 1.83~1.86;A260/A230 was 2.05~2.50),followed by the fast extraction(A260/A280 was 1.64~1.73;A260/A230 was 1.68~1.77)and the worst was SDS(A260/A280 was1.51~1.61;A260/A230 was1.56~1.76).The DNA was extracted by using improved CTAB method was suitable for PCR as a template,and successful for PAPD amplification.
